Abstract (EN)

Tunceli garlic (Allium tuncelianum KOLMANN) is a rare spread economically important endemic plant. The aim of this study was to micropropagate immature embryos of Tunceli garlic using. (i) 0.3, 0.5, 0.7, 0.9 and 1.2 mg/L 2,4-D, (ii) 0.1, 0.3, 0.5, 0.7 and 0.9 mg/L BAP, (iii) 0.1, 0.3, 0.5, 0.7 and 0.9 mg/L BAP + 0.5 mg/L NAA, (iv) 0.1, 0.3, 0.5, 0.7 and 0.9 mg/L BAP + 300 mg/L Putresin (v) 0.1, 0.3, 0.5, 0.7 and 0.9 mg/L BAP + 0.5 mg/l NAA + 300 mg/L Putresin. Calli formation rate of 2,4-D was compared to regeneration on other hormonal treatments. The highest callus formation (20%) was obtained on 0.3 and 1.2 mg/L treatment. The other variant (BAP, BAP + 0.5 mg/L NAA, BAP + 300 mg/L Putrescine, BAP + 300 mg/L Putrescine + 0.5 mg/L NAA) induced bulblets by direct somatic embryogenesis. Somatic embryos obtained on 2,4-D were transferred bulb growth medium (MS + 3% sucrose and 3% active carbon) followed by their transfer to soil.
