Evaluation of oxidative and nitrosative stress and antioxidant capacity in chi̇ldren with epilepsy

Abstract (EN)
Objective: The aim of this study was to compare nitrosative stress, total oxidant capacity (TAS), total antioxidant capacity (TOS), oxidative stress index (OSI) and 8-hydroxy 2-deoxyguanosine (8-OHdG) levels in epileptic patients receiving monotherapy and polytherapy with healty control group. The aim of this study is to discuss the relationship between antiepileptic therapy and free radical formation and oxidative damage in neurons. Methods: Between January 2019 - September 2019, 120 patients who followed in the Department of Pediatric Neurology, Faculty of Medicine, Gaziantep University, and 40 volunteers who applied to outpatient clinics of Pediatric Health and Diseases Department were included in the study. TOS, TAS, OSI, peroxynitrite and 8-OHdG parameters were studied in all groups. Results: The control group consisted of 19 girls (47.5%), 21 boys (52.5%), monotherapy groups consisted 30 girls (50%), 30 boys (50%) and polytherapy group consisted 31 girls (51.7%), 29 boys (49.3%). TOS and OSI in the polytherapy group was significantly higher than monotherapy and control groups (p=0.001). Peroxynitrite polytherapy group was significantly lower than monotherapy and control groups. Peroxynitrite was significantly higher in the monotherapy group compared to the control group (p=0,001). No difference between the groups of in the 8-OHdG parameter (p=0,929). Conclusion: As a result, parameter of TOS and OSI increased significantly in polytherapy group. This suggests that antiepileptic treatment increased oxidative stress. No difference between the groups in the 8-OHdG parameter suggested that the patients were treated effectively and DNA damage wasn't seen yet.
