Essential thrombocytosis in patients with bleeding and trombosis in the MPV and the relationship between JAK2 mutation

Aim: The frequency of JAK2 mutation in patients with essential thrombocytosis have been reported by 50-60%. About the relationship between tromboembolism and bleeding with JAK 2 situation in this patients, there are conflicting information available in the literature. There is no study which aims to determine the relationship between bleeding and trombotic events with the level of MPV in this group of patients. In our study we aimed to analyse relationship between Jak2 mutation frequency, bleeding and trombosis with MPV leves in essential thrombocytosis patients.Materials and Method: İn this clinical study, all patients files were scanned retrospectively who have a diagnosis ET and comes to İnonu Üniversity Medicine Faculty, TOTM Haematology Department between the date of 01.11.2000 and 01.03.2001. before the just beginning of study we take approval of the local ethic commity. We have obtained all datas, which are essential in our study, from files archieve of the hospital and automation system. In addition of standart chlinical and laboratory tests, all haematological, hystopatological, biyochemistrical, radiodiagnostical tests in the files were scanned carefully. All this datas of patients were memorized at the time of diagnosis. Patients file datas, trombosis and bleeding complications before the ET diagnosis, phisical examinations, chlinical and laboratory datas, radiodiagnostical tests were examined in detail. İf liver and spleen long axis was bigger than 150 mm with Abdominal USG, measure was accepted hepatomegaly or splenomegaly. Relationship of JAK2 mutation with age, gender, haemoglobin levels, haemotocrit levels, LDH levels, PCT, PLT, WBC, splenomegaly, hepatomegaly bleeding and thrombosis were examined. Also relationship between MPV with patients, who had a bleeding and thrombotic events history, examined in statistical analysis.Findings: Our study were included in the study of 102 patients diagnosed with ET. The frequency of JAK2 mutation in ET disease of our patients was found to be 55.1%. No relationship was found between JAK2 gen mutation and trombotic event but it was found a positive relationship between JAK2 mutation and bleeding complications. There was no relationship between JAK2 mutation and organomegaly in our study. Also no relationship was found between JAK2 frequency with gender and age. MPV values of patients, who had trombotic event or bleeding attack in history, did not differ significantly.Result: İn our study MPV values was not effected bleeding and thrombotic events in literature there are conflicting informations about relationship between JAK2 mutation positiveness with bleeding and thrombosis. İn order to resolve these contradictions, it is necessary to do more case including studies.
