Abstract (EN)

Anuk ÖO, Investigation of Tissue Protective Role and Phytochemical Composition of Chinar (Platanus orientalis L.) Leaf Infusion Against Ethanol-Induced Oxidative Stress In Rats, University of Van Yuzuncu Yil, Institute of Health Science, Faculty of Pharmacy, Departments of Basic Pharmaceutical Sciences, Biochemistry Science, Master Thesis, Van, 2018. Chinar (Platanus orientalis L.) plant is rich in kaempferol glycoside derivatives and is used in folk medicine against snake and scorpion poisoning, hoarseness, tooth and knee pain, wounds, inflammation and stomach discomfort however the effects of chinar leaves on liver and kidney organs are unknown. The aim of this study was to investigate the tissue protective role and phytochemical content of infusion obtained from dried chinar leaves against ethanol-induced oxidative stress in rats. After toxicity test, the infusion of Platanus orientalis (PO) were given at a dose of 20 mg/mL and 60 mg/mL for testing the biochemical changes, lipid peroxidation and antioxidant enzyme levels by ethanol induced oxidative stress in rats. In addition, silymarin known as liver-protective was used as positive control. Thirty male Wistar rats were divided into five groups which consist of Normal, Ethanol 20%, Ethanol 20%+Silymarin (10 mg/kg), Ethanol 20%+ PO-20 mg/mL infusion and Ethanol 20%+ PO-60 mg/mL infusion. According to the obtained in vitro results, benzaldehyde, palmitic acid, 2,4-Ditert-butylphenol, stearic acid, octadecanoic acid, linoleic acid and linolenic acid were determined in the GC-MS analysis in the extract. Additionally, the kaempferol and kaempferol derivatives were determined as dominant compound in the HPLC profile. In the Ethanol group, AST, ALT, LDH, GGT, uric acid and urea in serum and GST and MDA in liver and erythrocyte tissues showed a significant increase in levels compared to the Control group. AST, LDH, GGT, uric acid and LDL_C levels in serum and MDA (all tissues) significantly decreased in Ethanol+PO-60 mg / mL group according to Ethanol group. In addition, SOD, GPx and CAT activities in the kidney tissue of Ethanol group showed a significant decrease compared to the Control group and the GPx activity in all treatment groups increased significantly compared to the Ethanol group. These findings suggest that the administration of the determined infusion doses of the chinar leaves might have protective roles against liver and kidney damage induced oxidative stress by Ethanol in rats. Key Words: Chinar (Platanus orientalis L.), Silymarin, Antioxidant Enzymes and Oxidative Stress, Bioactive Compounds, Rat.
