Prof. Dr. Ahmet KAYRALDIZThis study was carried out to examine the chromosome breaks formed within humanchromosomes caused by mutagen Ethyl Methanesulfonate (EMS) and carcinogen MitomycinC (MMC), and to determine the chromosomes experiencing the most breaks. To achieve thisaim, the cells were treated with concentrations of 5x10-4 M, 10-3 M and 2x10-3 of EMS and0.0625μg/ml, 0.125μg/ml and 0.250μg/ml of MMC for 24 and 48hIn the study, the percentages of chromosome breakages which were observed withindifferent chromosomes in human peripheral blood lymphocytes treated with EMS for 24 and48 hours in both the control and the same dose were compared, and the difference was foundto be statistically significant. Moreover, the breakage percentages of the same chromosomeswere compared with the percentages of the control; and the difference was found to besignificant. That is, treatment with EMS led to more breakages. The most breakages viaEMS were seen in the chromosomes of 1, 2, 6, 4, X, 7, 3, 5, 9 and 8.The percentages of chromosome breakages which were also observed withindifferent chromosomes in human peripheral blood lymphocytes treated with MMC for 24and 48 hours in both the control and the same dose were compared, and the difference wasfound to be statistically significant. Moreover, the breakage percentages of the samechromosomes were compared with the percentages of the control; and the difference wasfound to be significant. That is, treatment with MMC led to more breakages. The mostbreakages via MMC were seen in the chromosomes of 1, 6, X, 4, 2, 3, 7, 9, 8, 5, 16, 10, 11and 13.Key words: Ethyl methanesulfonate (EMS), Mitomycin C (MMC), Lymphocyte cultures,Human chromosomes, Chromosome break.II
