Mouse embryonic fibroblast (MEF), The culture of cells is a powerful method for testing gene functions because of its easy accessibility, multiple growth rates, and multiple experiments. The high capacity of mouse embryonic fibroblast cells to transform into induced stem cells and their easy availability from embryonic tissues have paved the way for their therapeutic use. It is thought to be beneficial for drug discovery to be used in the treatment of many diseases. One of these diseases is disruptions in puberty. Kisspeptin and neurokinin protein family play an active role in the transition to puberty. In this study, the association of kisspeptin and neurokinin gene expressions of fibroblast cells obtained from mouse embryonic tissues with the number of passages was performed. Our study aims at a drug-free treatment with minimal immunoreaction using embryonic fibroblast cells with high capacity to induce pluripotent stem cells in patients with pubertal disorders. At the same time, we think that this method may contribute to the country's economy in terms of treatment method in the future. Since there are not many studies on the use of embryo fibroblast cells in the treatment of pubertal disorder and there are not enough resources, we believe that our study can contribute to filling this gap. In this study, embryonic fibroblast cell culture was established, the culture was maintained (passage) and Kisspeptin and neurokinin gene expressions were determined at different passage periods. Our aim is to determine kisspeptin and neurokinin expressions in MEFs with high capacity to transform into pluripotent stem cells and to investigate their suitability for stem cell-based treatment of pubertal diseases. Expression of Kisspeptin and Neurokinin could not be detected in MEFs, but we found that both directionality receptors KISS1R and TACR2 receptors were expressed and this expression decreased depending on the number of passages.
