Investigation of the effects on microleakage of ozone gas and traditional cavity disinfectant at teeth which are restorated with different restorative materials

Aim: The aim of this in-vitro study is to investigate the effects of ozone and conventional cavity disinfectant on microleakage at the cavities which are restorated with different restorative materials. Material and Method: 90 human molar teeth extracted for various reasons were used in this study. Teeth are examined carefully one by one and cared not to have caries, restorations and fracture. And then soft tissues are scaled with a scaler and polished with pumice and polishing plastic.The teeth are kept in distilled water at room temperature untill study is done. Standard Black V cavities are prepared on buccal surfaces of all teeth with a cylinderyque diamond bur (Plus, BR31B, P.R.C) with air and water cooler. All cavities are prepared 3 mm mesio-distal size, 2 mm occlusal-gingival size and 1,5 mm depth. All cavity surfaces are bisotated 0,5 mm wide. Teeth are divided to 3 main groups which contains 30 teeth. Group 1;ozone applied for 6 seconds, Group 2; %2 clothexidine glucontae applied, Group 3; any disinfection is not applied. And then all groupes are divided 3 sub-groupes which contains 10 teeth. All sub-groupes are restorated with silorane, ormocer and traditional composite according to the producter's directions. All specimens are incubated for 24 hours at 37'°C (Nüve Incubator EN Ankara,Turkey) before the finishing and polishing procedures. Aluminum coated discs (So-Flex,3M ESPE,St. Paul,MN,USA) are used for finishing and polishing. After incubation process, all specimens are thermal cycled at 5± 2 ºC and 55 ± 2 ºC 1500 times (for 30 seconds). And then apical foramens of the teeth were sealed with flow composite and specimens were coated with acid resistant nail polish 1 mm away from the restoration border. All specimens were incubated in basic fucsine at 37'°C for 24 hours (Nüve Incubator EN 500 Ankara,Turkey). And then the teeth are washed under water to move excess paint. The teeth were seperated with 0,2 mm thickness diamond seperator (Horico, Diamond Instruments, Germany) and tribune (DEGA z, ceramik, Made in P.R.C) buccal-lingual/palatinal direction with water cooler. Sections were examined under a microscope Occlusal and gingival microleakage values of slice surfaces were obsereved at x57 magnification under stereooptical microscope (Leica Z16 APO, Germany) by a seperate researcher and scores were recorded. Gingival and occlusal microleakage degrees were scored between 0-4 by a standard scale.1 tooth was choosen from each group randomly. And then the surface morphology between resine-tooth hard tissue of choosen teeth were observed under scanning electron microscope (FEI, Quanta FEG 250, Germany) at various magnifications and photographed. Statistically results were reviewed with ANOVA and Kruskal-Wallies Tests. Results: Statistically significant difference of microleakage between cavity disinfectant and control groups' occlusal and gingival lines was not found (p>0,05). There was no differences at occlusal lines of composite subgroups (p>0,05) but also significant difference at gingival lines was found (p<0,05). The most succcessful results at gingival lines were found with silorane. Conclusion: In our study, we found that there is no effect of cavity disinfectant to microleakage at occlusal and gingival line. Statistically significant difference was found among the composite materials, between clorhexidine and control groups' gingival lines.The most successful results at gingival lines was found with filtek Silorane.
