Phylogeny analyses of retrotransposon genes in common bean (Phaseolus vulgaris L.)

Abstract (EN)

In phylogeny analysis, retrotransposon based moleculer marker system efficiently used for identification of genetic resources. In this study, 67 common bean genotypes from different origins in Turkey and the world by LTR retrotransposon based inter-Primer Binding Site (iPBS) marker system, has been identified at the molecular level. In iPBS marker systems, 180 polymorphic bands were obtained by 47 primers. According to the result, the highest and lowest PIC value determined as 0.94 (primer iPBS-2394) and 0.03 (primer iPBS-2375), respectively. Population structure was determined using STRUCTURE software and mainly two groups (K=2) were identified among 67 common bean genotypes. Genotypes in separated two main groups, referred to as a mixture, were determined. In the analysis of molecular data, NTSYS program obtained Jaccard (J)'s similarity coefficient that was compared with genetic similarity dendrogram and parallel results were observed. Group 1: mainly belong to Turkey location genotypes, Group 2: genotypes belonging to different countries and Group 3: two genotypes of India. In iPBS marker analyses, the genetic similarity ratio between different genotypes varied between 0.14 to 0.99. The closest genotypes was determined as (15) Bandırma/Turkey (Yellow sugar) and (42) Turkey (Volare) and the similarity value was established as 0.99. The furthest genotypes was determined as (9) Bozdağ/Turkey (Kula barbunya) and (52) India (Dolic hos), and the similarity value observed as 0.09.
