Özet (EN)
Recently, interaction of transition metal complexes with nucleic acids have gained much attention due to their great importance in the design of chemotherapeutic agents. There are transition metals in biological systems and metal ions present in synthesized complexes were known to accelerate drug efficiency. Thus, it is not suprising that transition metal complexes to evaluation as potential drugs. DNA is an important celular reseptor and DNA is a primary intracellular target for many antiviral and anticancer therapies. Therefore, desinging and development of new molecules capable of binding to DNA or cleavage to DNA is required. When biological properties are taken into account, transition metal complexes are the potential agents for the development of new drugs in the struggle against diseases. Hence, investigation of the interaction of transition metals and their complexes with DNA is quite important. In this study, interaction of benzimidazole derivatives complexes containing ferrocene unit complexes with DNA were examined by UV-Vis absorption titrations and thermal denaturation methods. These assays demonstrated that complexes were localized by intercalation between the base pairs of DNA. Using agarose gel electrophoresis, it was determined that complexes of hydrolytic activity was at pH 7 and it was identified how to affect the concentration of the nuclease activity. Remarkably increased activity was determined in cleavage experiments performed in the presence of various activators such as H2O2 (0.4 M), ascorbic acid (2.5 mM) and mercaptoethanol (0.4 M) The antioxidant activities were performed by 2,2-Diphenyl-2-picryl hydrazyl (DPPH) radical scavenging and superoxide dismutase (SOD) activity tests. Especially complex 1 (K1), all complexes have exhibited good free radical scavenging activity.
