ABSTRACT Comparison of Some of the Cytosolie and Mitochondrial Enzyme Activities which Play Rote in Fertile and Infertile Semen Samples It is reported that infertility is due to male factor approximately at 50%. Spermogram too important in the evaluation of male fertilization potential. So sperm can be evaluated according to concentration, motility and morphology. In this study, in fertile and infertile semen groups, Malate dehydrogenase (MD1 1 ) and lactate Dehydrogenase (LDH) activities (to study energy metabolism of sperm) and levels of glucose, fructose and reduced glutathione (GSM) were investigated. Free testosterone and teptin levels were compared to explore the efficiency of local hormones. Infertile group was classified in to four subgroups according to concentration, motility and morphology. Enzyme activities were measured with kinetic method, GSH, fructose and glucose levels were evaluated spectrophotometrically and hormone levels were measured with R1A method. NAD-MDH (the key enzyme of energy metabolism) and LDH activities and GSH levels were found significantly (p<0.05) higher in seminal plasma of fertil group. We believe that these variables may have an important contribution in the evaluation of sperm functions. In seminal plasma of oligoteratospermic group, levels of free testosterone and leptin were found significantly lower (p<0.05) than control group. Â positive correlation was found between testosterone level and seminal plasma NAD-MDH, so it is thought that testosterone may be a factor that regulates activity of this enzyme. It is reported that in addition to fructose sperm also use glucose, and consumption of these sugars similar to each other. Fertile group use more fractose and glucose than infertile group. It is showed that glucose is important for energy requirement of sperm. We reported that NAD-MDH works better than NADP form, both in seminal plasma and sperm. It is found that LBH activities of all groups were significantly (p<0.05) lower than control group. Keywords; Lactate Dehydrogenase, teptin, Malate Dehydrogenase, tern testosterone. XIV
