Abstract (EN)
Food additives are commonly used in the food industry for improving the durability, protecting the nutritional value, determining the physical structure, flavor, odor of the foods. The presence of several artificial food additives synthesized by chemical methods in foods and beverages causes insecurity in consumers. Considering losses of food additives; amount of food additives in foods (WHO and the amount of permitted by the Turkish Food Codex) constitute importance for the consumer's health. Analytical methods are required for the identification and quantification of these additives. These requirements are increasing simultaneously with the developing technology. In this context, various methods are used in the industry to determine the amount of additives in foods and the studies in this direction are increasing day by day. The accuracy, selectivity, time and cost are very important for the used method when considered in terms of consumer and industry. In this thesis work, UHPLC method (simple, short term analysis, valid and reliable) developed for the simultaneously determination of acesulfame K, aspartame, caffeine, sodium benzoate additives found in sodas. Validation studies of developed method the were performed and applicability of the analysis was shown for the all the sodas. Also, method was announced because it does'nt need any pre-separation or derivatization process, includes a minimum of preparation steps and time, causes minimum chemical xviii consumption. Developed and validated UHPLC method was applied to other sodas by the market and obtained the results confirmed the success of the method. Developed and validated UHPLC method was applied to the HPLC system and the results obtained for both systems were compared. UHPLC system was demonstrated more advantageous than HPLC system and the system selection suitability of the UHPLC was determined. The most important advantage of the UHPLC method requires short analysis time and less solvent. Solvent consumption was determined to decrease to 80% as a result of research conducted in the literature. Analysis time and reduction of solvent consumption are important in terms of low cost for many laboratories. In addition, analysis of many studies conducted for method development with UHPLC in the literature have been performed as soon as possible and the method has been expressed that less solvent and time required according to the HPLC in adverse conditions have been described only as it can influence the direction of reducing the column life of the high pressure in the UHPLC system. Within this thesis work, simple, low-cost, short analysis time, low analyte consumption, high selectivity, reliable UHPLC analysis method was developed for simultaneously detecting additives in various sodas and method previsualized to suitability for quantitative analysis. Average recovery values obtained in the validation study is 98.88% for Acesulfame K, 98.99% for Aspartame, 99.36% for Caffeine, 98.93% for Sodium Benzoate respectively. Correlation coefficient (r2 ) obtained in the linearity study is 0.9999 for Acesulfame K, 0.9996 for Aspartame, 0.9994 for Caffeine, 0.9998 for Sodium Benzoate. Calculated LOD value is 1,6 µg/mL for Acesulfame K, 4,3 µg/mL for Aspartame, 3,9 µg/mL for Caffeine, 4,1 µg/mL for Sodium Benzoate. The calculated LOQ value is 5,2 µg/mL for Acesulfame K, 14,4 µg/mL for Aspartame, 12,9 µg/mL for Caffeine, 13,7 µg/mL for Sodium Benzoate
