Detection of genotoxic effects of food additive sodium metabisulfite using RAPD/PCR method

Abstract (EN)

In the present study, the RAPD-PCR assay was used to evaluate the possible genotoxic effects in the genomic DNA of human peripheral lymphocytes cells that exposed with 75 µg/ml, 150 µg/ml, and 300 µg/ml SMB concentrations for 24 and 48 hours treatment periods. For this purpose, 13 primers consisted of 10-mer were used in this study. The genotoxic effect of SMB was evaluated according to band newly formed or absent bands compared with control bands in the RAPD band profiles. Then the values of GTS were calculated of sample of each primer. SMB produced polymorphic bands all on concentrations of 24 and 48 hours when compared with control bands. SMB significantly changed the value of GTS compared with control for at all concentrations. The genotoxic effect as a result the treatments of the lowest two doses of SMB was determined almost positive control proceedings genotoxic effects of sodium azide (SA) is so. Morever, the genotoxic effects resulting from treatment of the highest concentration of SMB was detected that SA has demonstrated the genotoxic effect was found to be more.
