Comparison of different diluent used in turkey (Meleagris gallopavo) semen freezing

Abstract (EN)

This study was conducted to improve the development of long-term preservation methods of turkey semen using different extenders. Semen was collected from five turkeys by manual massage method twice a week and a total of 44 times throughout the study. The means amount of the resultant nativ semen ejakulate volume (ml), sperm concentration (x109 sp/ml), motility (%) and vitality (%) were recorded as 0,22±0,01 ml, 3,5±0,17 x109 sp/ml, % 77,0±1,44 and % 86,2±0,95 respectively. In this study, used Glucose, Tris-Glucose, Lactated Ringer and Lactated Ringer-Glucose extenders containing % 5 DMSO cryoprotectant. The semen collected from turkeys combained and divided into four equal parts. The semens equilibrated at 4°C for 90 min after diluted 1:3 with extenders. The semens stored at -196 o C on liquid nitrogen after frozen at -80°C for 5 min on liquid nitrogen vapour. After post-thaw motility for Glucose, Tris-Glucose, Lactated Ringer and Lactated Ringer-Glucose detected as % 43,3±1,62, % 12,2±0,66, % 12,6±0,92 and % 24,6±1,53 and for vitality detected as % 55,8±1,89, % 23,8±1,58, % 21,5±1,10 and % 36,5±1,59 respectively. According to the findings obtained in this study, Glucose extenders was found to be more effective in the long-term preservation of turkey semen than the others (p<0.01).
