Assessment of the effects of oxolamine in an experimental peritoneal injury model induced by hypertonic dialysate solution

Abstract (EN)

The aim of this study is to investigate the effects of an anti inflammatory matter which is oxolamine in a model of peritoneal injury induced by hypertonic dialysis solution in rats. 21 Wistar Albino rat ranging in age from 8 to 10 weeks and ranging in weight from 200 to 250 grams were included in this study. They were divided into 3 groups such as control group (n=7), dextrose group (n=7) and dextrose+oxolamine group (n=7). To control group, saline was given intraperitoneally in a dose of 10 ml every 28 days. To the dextrose group, 10 ml %3.86 glucose dialysis solution was given intraperitoneally every 28 days. To the oxolamine group 10 ml %3.86 glucose dialysis solution was given intraperitoneally every 28 days. With these applications, oxolamine was given as 50 mg/kg/day with gavage every 28 days. Solutions were given intraperitoneally once a day using insulin syringe respectively. At the end of four weeks parietal peritoneal sections of rats were dyed with H-E and peritoneal thickness, inflammation and fibrosis were viewed under light microscope. Additionally, transforming growth factor beta, connecitve tissue growth factor and fibronectin were investigated. In the assesment with the H-E dye, peritoneal thickness and inflammatory cell infiltration were incresed in the dextrose group. Immunohistochemically, peritoneal TGF-β, CTGF and fibronectin levels were high in dextrose group. However TGF-β, CTGF and fibronectin levels were low in oxolamine group as compared to dextrose group. With the H-E dye we concluded that oxolamine decreases the peritoneal injury induced by hypertonic dialysis solution. Additionally to the anti inflammatory properties of oxolamine, it may help to prevent peritoneal thickening by decreasing TGF-β, CTGF and fibronectin. Positive effects of oxolamine on preventing peritoneal fibrosis were determined and it was found to have some roles on growth factor pathways related to fibrosis.
