Abstract (EN)

L-glutaminase (L-glutamine amidohydrolase EC 3.5.1.2) is the important enzyme that catalyze the deamination of L-glutamine to L-glutamic acid and ammonium ions. It has an important role in the nitrogen metabolism. Recently, L-glutaminase has received much attention with respect to its therapeutic and industrial applications. Beside it acts as potent antileukaemic agent and it shows flavorenhancing agent in the production of fermented foods, too.Glutaminase activity is widely distributed in plants, animal tissues and microorganisms including bacteria, yeast and fungi.It has aimed that microbial production of glutaminase enzyme from Hypocrea jecorina pure culture and Trichoderma reesei 1A mutant strain, determination of optimum conditions and calculation of kinetic parameters of these produced enzymes. As a result of work, it was determined optimum pH value as 8.0 and optimum temperature as 50oC for glutaminase enzyme produced both microorganism species. KM and values, the kinetic parameters of enzyme produced Hypocrea jecorina pure culture and Trichoderma reesei 1A mutant strain, were determined as 0.491 mM, 0.647 mM of KM values and 13.87 U/mL, 13.01 U/mL of Vmax values by plotted Lineweaver-Burk graphs, respectively.In addition, termal stability and storage stability of glutaminase enzymes produced from both microorganism species were determined as very high.
