Comparison of protective effect of rosuvastatin with ischemic preconditioning on ischemia reperfusion injury, on in vivo rat hearts.

Oxidative stress due to reperfusion after ischemia causes severe functional and structural damage. Free oxygen radicals are responsible for this damage. Besides techniques such as ischemic preconditioning the protective effects of many antioxidant substances has been shown in many of the lately carried out studies.Rosuvastatin -one of the statin compounds- by its pleiotropic effects, found useful in decreasing cardiac damage by antioxidant and anti-inflammatory effects watched in reperfusion stage.In this study, protective effect of pharmacologic preconditioning with rosuvastatin compared with ischemic preconditioning on hearts exposed myocardial ischemia, by occlusion of left coronary artery. Male wistar albino rats used in this study split into four groups; Sham operated group, Ischemia Reperfusion group (30 min ischemia, 120 min reperfusion), Ischemic Preconditioning group (5 min ischemia followed by 5 min reperfusion; 3 sets) and Pharmacologic Preconditioning (10 mg/kg rosuvastatin, IV infusion). Blood samples gathered from all groups on the 10th min of stabiliszation and are tested for creatine kinase-myocardial band (CK-MB) values and blood taken replaced with same amount of serum physiologic in body temperature. During all protocol, tension arterial and heart beat count tested three times on 10th min of stabilization, on 60th, 120th mins of reperfusion protocol. At the end of the experiment all hearts are stained with Evans blue and 2,3,5-Triphenyltetrazolium cloride (TTC) stain to determine the necrosis field.At the end of the experiments, CK-MB values showed a significant decrease in at the end of the ischemia and 60th 120th mins (p<0.05) of reperfusion. Furthermore, decrease in CK-MB levels in pharmacologic preconditioning group compared to ischemic preconditioning groups was significant (p<0,05). Comparison done for change in necrosis field, ischemic preconditioning group showed lower values for both mass and volume (p<0.01) compared to ischemia reperfusion group. Necrosis area was smallest in pharmacologic preconditioning group (p<0,01).As a result pharmacologic preconditioning with rosuvastatin is more protective and effective compared to ischemic preconditioning against ischemia reperfusion injury.
