Using new generation tissue culture systems in in vitro clonal banana production

In this study, solid culture method and Plantform and Setis temporary immersion bioreactor systems were used comparatively in order to propagation, rooting and acclimatization of Grande Naine and Azman banana cultivars for rapid, cheap and mass production in in vitro conditions. Micropropagation rate, plant height, number of leaves, fresh and dry weight parameters were investigated in the micropropagation stage across 8 subcultures. Rooting rate, plant height, number of leaves, number of roots, root length, fresh and dry weight parameters were investigated in the rooting stage. Photosynthetic pigment analyzes and stoma examinations were performed throughout all stage. In the micropropagation stage; 20% increase in Plantform system, 12% increase in Setis system in Grand Naine, 82% increase in Plantform system and 98% increase in Setis system in Azman were determined compared to solid culture. At the rooting stage, higher data were obtained from bioreactor systems than from solid culture. Plants coming from bioreactor systems acclimatizatied faster and developed healthier in the greenhouse stage. It was determined that stomata were more activeand pigment accumulation was higher in bioreactor systems. One of the most important issues in banana clonal propagation is genetic variations occurring across subcultures. Leaf samples were taken from each system and variation in plants was investigated using SSR markers. No variation was observed from the initial stage to the greenhouse stage. As a result, it has been determined that bioreactor systems are an important alternative for mass production of bananas.
