Lung cancer is the most common disease with the highest incidence and mortality rate. Many years of studies have failed in most lung cancer treatments. In this study; examination of the expression profiles of genes involved in the apoptosis pathway induced by the TNF-related apoptosis-inducing ligand (TRAIL) and the genes related to these genes in A549 cells. The aim of this study was to determine how various combinations of recombinant TRAIL (rTRAIL) and Bortezomib antitumor agents affected this pathway gene expression and protein quantities. For this purpose, expression and protein profiles of TRAIL-related apoptosis pathways were investigated which is plays important role on pathogenesis of lung cancer. Also effects of antitumoral agents TRAIL and Bortezomib has been investigated on apoptosis pathway. Human lung cancer cell line (A549) cell cultures was make fora aim of the study and, the cell cultures divided into 3 groups; first group is control, TRAIL protein applied externally to the second group, TRAIL protein and Bortezomib applied to the third group. Then, protein and expression levels of genes, which are functionary in apoptosis pathway, determined by Western Blot method. Ratio of apoptosis controlled by apoptosis assays. The aim of study is to acquire more information about effect of apoptosis pathway dependent TRAIL induced on obtained data in emphysema and lung cancer and effects of antitumoral agents on apoptosis pathways. In our study, no significant statistical difference were observed between the control and treatment groups for TRAIL, TRAIL- R1, TRAIL-R2, TRAIL-R3, TRAIL-R4, OPG, Bcl-2, Bax, Bcl-XL gene and protein expressions.
