The investigation of some probiotic properties of human resources vagina lactic acid bacteria

Abstract (EN)
In this study,the isolated Lactic acid bacteria (LAB) which were gathered by taking swab samples from the vaginal areas of 60 healthy women who applied to Kırşehir Ahi Evran University Training and Research Hospital, who were between the ages of 18 and 45, who did not go through menopause, who did not carry out birth control with any contraceptive method, and had not use antibiotics within 3 months were used. As a result of the identification that was done according to 16S rDNA sequence analysis, 7 different species were obtained from 20 selected isolates. In our study, L. plantarum was the most frequently seen species with the rate of %25, and this strain was followed by the strains of L. rhamnosus and L. paracasei with 20%, by P. acidilacitici with 15%, and by L. gasseri, L. acidophilus, L. crispatus and Lactobacillus spp. with %5. Various probiotic properties of the identified strains were investigated at in vitro conditions. At different pH conditions (2.0, 2.5 and 3.0), the strains that were resistant to the acidity at the highest level were identified as P. acidilactici L6, L. rhamnosus L8, L. plantarum L9, P. acidophilus L11, L. rhamnosus L15, and L. plantarum L18. L. plantarum L9, L. plantarum L16, L. plantarum L18, L. plantarum L19 and L. plantarum L21 strains were able to protect their vitality in high bile salt concentrations (0.3%, 0.5%, and 1.0%). High levels of resistance were determined in antimicrobial activities of strains, in particular for the antibiotics that are used in therapy; and against gentamicin, tobramycin and amikacin antibiotics all strains were found to be resistant, while they were found to be 80% resistant to vancomycin and teicoplanin antibiotics. 19 different pathogenic microorganisms were used in order to determine antagonistic activities of LAB, and it was determined that strains generally have an inhibitory effect against pathogens. In the study that was carried out to determine the cholesterol assimilation capacities of the strains, it was determined that the strains that assimilate cholesterol in the highest rate were L. gasseri L10 and L. paracasei L20 strains with the rate of 39.65%. In the autoaggregation study, at the end of the 4th hour, L. plantarum L16 strain was found to have the highest autoaggregation capacity with the rate of 85.2%. In the coaggregation study, it was determined that the strain that makes the best coaggregation with E. coli ATCC 25922 was L. plantarum L9 with the rate of 47.32%, and the strain that makes the best coaggregation with C. albicans ATCC 10231 is the L5 strain with the rate of 52.05%. In the study that the antiproliferative effect of vaginal LAB on HeLa (human cervical cancer cell line) and Caco-2 (Human Colon Carcinoma Cell Line) cancer cell line was identified, it was observed that L. paracasei L1, L. rhamnosus L8, L. rhamnosus L12, L. rhamnosus L13 and L. plantarum L19 strains create an important antiproliferative effect. As a result, it is considered that most of the isolated strains have probiotic potential; and that, L. plantarum L9 and L16 strains, alone or in combination with other strains of Lactobacillus, are the potential candidates that can be used for the production of oral or vaginal preparations.
