Researching the efficency of intraperitoneally injected Melatonin and Pinealectomy in experimental model of acute pancreatitis

The acute pancreatitis is a serious illness that progresses with high morbidity and mortality with damaging of pancreas tissue due to biliary causes as a result of inflamation, infection and serially physiopathological event, among emergency surgical diseases.A lot of clinical and experimental researches are made to explain this complex physiopathological event and to introduce endocrinologic, enzymatic, biochemical and histopathological changes. In consequence of these studies, the changes in the cellular level that performs in the physiopathological and clinical lots of mechanism of reaction are explained. However, due to unknown mechanism about this subject, pancreas and pancreaticobiliary system functions and diseases, are still open to observation with experimental researches and progressed techniques.In this study, we purposed to introduce decrease or increase in the level of melatonin in blood of experimental model of obstructive pancreatitis on rats, or the effects on the physiopathological events of pancreatitis which changes are caused histopathological and biochemical in pancreas tissue. With this purpose, the rats with acute pancreatitis whose blood melatonin level are different are occured by applying pinealectomy in order to remove endogenous melatonin source or by using exogenous melatonin injection. Acute pancreatitis subjects (rats) whose melatonin levels are different, histopathological changes in white blood cell infiltration, edema and haemorrhage in tissue, degeneration of acinar cells and apoptosis are observed. Besides, biochemically aspartate transaminase (AST), alanin transferase (ALT), amylase, lactat dehydrogenase (LDH) and bilirubin values with malonyldialdehyde in tissue (MDA), glutathione (GSH) and superoxide dismutase (SOD) levels are observed. According to this parameters, differences between the groups are considered and its effects on melatonin acute pancreatitis are explicated.In the subjects which have acute pancreatitis with pinealectomy or without pinealectomy who are intraperitoneally injected 0,1 ml melatonin (20 mg/ml), decreases in polimorf nuclear leukocytes infiltration, edema, haemorrhage and acinar cell necrosis, apoptotic cell number are observed. Besides, decrease in the values of AST, Amylase, LDH, Bilirubin and MDA level of tissue are determined.As a result, in this study, it is concluded that exogenous melatonin injection on acute pancreatitis have an important role on lipid peroxidation products and free oxygen radicals causes inflamatory reaction in tissue and helps to healing by decreasing oxidative injuries and reducing levels of MDA, AST, Amylase, LDH and Bilirubin.Keywords: Acute Pancreatitis, Melatonin, Experimental Study
