ABSTRACT MASTER THESIS EVALUATION OF TOTAL OXIDANT TOTAL / ANTIOXIDANT LEVELS IN PATIENTS WITH INTRACEREBRAL BLEEDING SERCAN KALKAN YOZGAT BOZOK UNIVERSITY SCHOOL OF GRADUATE STUDIES MEDİCAL BIOCHEMISTRY SUPERVISOR: PROF. DR. MUHAMMED FEVZİ POLAT SUPERVISOR: DOÇ. DR. ALPER GÜMÜŞ Objective: The accumulation of arterial or venous blood in the brain's parenchym or ventricular system is called intracerebral bleeding (ICD). As you age, the risk increases and it is more common in men than in women. Research has shown that women are more at risk of dying and dependent in the early universe. According to the study, the incidence of ischemic stroke was 71.2%, while the rate of intracerebral bleeding was 28.8%. The most common risk factor for IBS is hypertension. Hypertension causes disturbances in the small penetrating arteries in the brain, fractures in the flat muscles in the arterial wall, and fibrinoid necrosis. These degenerative disorders are seen in medium and small arterioles. An acute rise in arterial blood pressure (ACD) causes IBS. The fact that AKB is high suggests that the capillaries can cause bleeding from the veins and arterioles. In the majority of cases, chronic changes in the cerebral veins are pathophysiological events that cause the formation of ICH. As a result of the thickening of chronic HT arteries and small vessels, blood flows into the brain tissue and causes the formation of hematomas and mechanical damage to the surrounding tissues. By comparing oxygen (TOS) and antioxidant (TAS) levels, it is necessary to understand the importance of associating them with brain bleeding. Method: A study group was formed for 46 patients who were diagnosed with intracerebral hemorrhage after a detailed examination by the specialist physicians who applied to the Brain Nerve Surgery Clinic and the intensive care unit of Başakşehir Çam and Sakura City Hospital with complaints of headache, high blood pressure, and nausea. In the control group, 47 patients from other patient groups who were not diagnosed with intracerebral hemorrhage who applied to the Neurosurgery clinic were included. Patients under the age of 18 and over the age of 65 will not be included in the study. The excess blood samples taken from the peripheral veins of these patients were first allowed to clot for 20-30 minutes at room temperature, then centrifuged at 4000 rpm for 10 minutes, and the serums separated into eppendorfs were stored at -80 degrees. At the beginning of the test, the samples were brought to room temperature. TOS and TAS kit were applied according to the test principle. The kit materials were used in accordance with the steps specified in the booklet, and the color change rate that occurred after the substrate reacted with the enzyme bound to the antibody was measured in the spectrophotometer, and TOS and TAS results were found. Statistical analyzes were calculated with SPSS 22.0. Calculated results are given as mean and standard deviation (+/-). The results were evaluated with the Shapiro Wilk measurement method, and normal values were evaluated using parametric (t-test) and non-normal values were evaluated using non-parametric tests (Man Whitney U test and KW kruskal wallis) and power analysis was repeated at the end of the study. Results: According to the study, TOS, TAS and age values were evaluated statistically. TAS values were found to be higher in the control group than in the patient group. No statistically significant difference was detected. Although TAS values by gender were higher for men than for women, the statistical difference did not yield significant results. It was found that those under 45 years old were higher than those over 45 years old. No statistically significant difference was detected. TOS values were found to be higher in the patient group than in the control group. Statistically significant difference was detected. Although the OSI values by gender were higher in women than in men, no statistically significant difference was found. When TOS and OSI of age groups are evaluated under 45 years old and over 45 years old; It was found that over 45 years old is high. No statistically significant difference was detected. Conclusion: The fact that serum TOS and TAS values and, as a result, OSI levels are high in patients with ICH disease, suggests that they may be related to these diseases.
