Prof. Zahid Saadoun AZ˙IZ Tuberculosis (TB) is an important health problem as a life-threatening infectious disease all over the world. Approximately 30% of the world's population is infected with Mycobacterium tuberculosis. According to the World Health Organization (WHO) 2020 report, it was reported that approximately 500,000 people developed rifampicin-resistant tuberculosis in 2019, and 78% of them were multidrug-resistant tuberculosis. 10 million people contracted TB disease in 2019. Therefore, early diagnosis of infection is very important in terms of treatment. Today, the development of DNA-based assays for the detection of Mycobacterium tuberculosis complex has significantly increased sensitivity compared to traditional microscopy and culture. In this study, it was aimed to reveal the isolation and molecular diagnosis of Mycobacterium tuberculosis with Genexpert MTB/RIF and TB-LAMP techniques in patients with pulmonary and extrapulmonary tuberculosis in Iraq. 141 specimens such as sputum and body fluid samples were taken from the patients who applied to the National Respiratory Diseases Center from central, southern and western parts of Iraq. Routine acid-fast bacilli test was performed on the samples. As a result of the test, 58 (54 pulmonary + 4 extrapulmonary) samples were determined to be positive. Genexpert MTB/RIF and TB-LAMP techniques were used for molecular diagnosis. The age groups included in this study were between 6 and 65 years old. Men were affected 70 (78.77%) more than 20 (22.22%) women. From a total of 107 lung samples, 76 samples were diagnosed as positive. Extrapulmonary 14 samples were diagnosed as positive. Rifampin resistance was diagnosed in two cases. In addition, the molecular diagnosis of the same samples was made with the TB-LAMP technique, and 70 samples were positive for pulmonary tuberculosis and 8 samples from extra pulmonary samples were diagnosed as positive. As a result of the study, the Genexpert MTB / RIF technique has shown that it has more specificity and sensitivity in diagnosis.
