Abstract (EN)
Heart failure is an important health problem in the world, its prevelance on the increases. İschemic cardiomyopathy is defined as EF < %40 with myocardiyal wall motion abnormality due to coronary artery disease. İt is an important cause of morbidity and mortality. Atherosclerosis is the most common reason of Ischemic heart disease, Coronary artery calsification (CAC) is associated with coronary atherosclerosis. There is a significant relation between coronary artery wall calsification and coronary artery disease(CAD). The measure of coronary artery calsification is an indicator of subclinical atherosclerosis . İn some studies it has been showed that independent from convantional cardiovascular risks , coronary artery calsification suggests data about prognosis. Matrix Gla protein (MGP) appears to be an important protective modulator against calsification since it is a potential inhibitor of tissue calcification. Therefore , MGP as an inhibitor of calsification recently has been more important in studies. In this respect, it is aimed to establish the relationship between the distributions of nucleotide alterations found in promoter and coding regions of MGP gene in İCM patients and DCM patients. DNA samples were obtained from 49 patients with İCM and 74 patients with DCM (Totally from 123 patients). The DNA samples obtained were analyzed by Polymerase Chain Reaction (PCR) method using the 2 sets of primer pairs which cover the coding (Thr83Ala in exon 4) and promoter regions (T-138C ) of MGP gene. Amplified regions were analyzed by Restriction Fragment Length Polymorphism (RFLP) method for possible polymorphisms.In the chi-square analysis for T-138C gene polymorphism there is a statistical significant difference between genotypes(p=0,019) and allels(p=0,011). But in analyses for Thr83Ala gene polymorphism there is not difference between groups.The results of this study demonstrated that there is a significant relation between the T-138C gene polymorphism and ICM.Keywords: İschemic Cardiomyopathy, Dilate Cardiomyopathy, Coronary Artery Disease, MGP Gene Polymorphism
