Micropropogation of jumbo and chester thornless blackberry cultivars under in vitro conditions

Abstract (EN)

Rubus species having heterozygotic characteristics and when it is propagated by seeds the seedlings shows varition. As a result of this, generatif propagation method is not suitable for produce truth the cultivar name in blackberry propagation. For this purpose, micropropagation methods are become very important and having many advantages compare to the classical propagation methods such as cutting, stip layring etc. In Turkey, blackberries are propagated using cutting methods especially softwood and wood stem. However, using cutting propogation methods have several disadvantages. The most important problem is rooting especially in Chester Thornless variety. Addition to this, limited plants can be propogated using classical propogation method and thus plant demand of growers can not be covered. The scope of this thesis is to investigate in vitro propagation of ``Jumbo?? and ``Chester?? Thorrnless blackberry cultivars and to set in vitro propogation methods. For this purpose, we aimed to conduct suitable surface steriliziation methods, shooting, multiplication and rooting mediums using various basic nutrient mediums (Woody Plant Medium (WPM, after Lloyd & McCown and plant growth regulators), BAP (1.5, 2, 3 mg/l), NAA (0, 0.1, 0.2, 0.4 mg/l), GA3 (0, 0.25, 0.50 mg/l), IBA (0,0.1,0.2,0.4 mg/l). The highest number of shoots was from the media that contain 2.00 mg/l BAP and the highest number of roots were obtained from 0.4 mg/l NAA. When NAA and GA3 added to the media a multiplication rate was decreased
