Abstract (EN)

In this study, we investigated the cytotoxic effect of phosphine compounds on K562 cell line that originated by blastic phase of cronic myeloid leukemia. The possible cytotoxic effect of new synthesized organophosphine compounds on the some cancer cells especially K562 cell line could be the first step of anticancer drug investigation.Before starting this study, K562 cell line was passaged. The 104 K562 cells was seed out each well of 12-well-culture plate. Two well determined as a blank and was put medium and DMSO. Organophosphine compounds were added each wells five different concentrations (1ng/mL, 10-1 ng/mL, 10-2ng/mL, 10-3ng/mL and 10-4ng/mL). MTT (3-[4,5-Dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide; Thiazolyl blue was added each wells of 96-well-culture plate and incubated four hours. After mixing by using vortex, finally DMSO was added each wells. Microplate plates were incubated 10 minutes in the CO2 incubator and after the plates were evaluated by microplate reader on the 545 nm and were determined the absorbance values. The results were analysed by statistically methods.The only one of the three new synthesized organophosphine compounds had cytotoxic effect by used MTT test. In this study, we showed that the possible anti cancer effect of the [RhCl(Ph2PCH2)2NCH3]2 might be give inspiration to us with hope in the experimental animal models.
