Enzymes that are being used in almost all areas in industry is usually obtained from microorganizms. Because, enzymes are produced by microorganizms have some advantages when compared with enzymes produced by plants or animals, they have considerably higher catalytic activity, they don?t from undesirable by-products, they are more stable and relatively cheap, and they can be obtained much quantity.Proteases that are a group of indüstrial enzymes have an important role. Proteases were used detergent, pharmaceutical, leather, meat, milk, beer indüstries, hydrolysis of proteins such as horn, feather and hair, recovery of silver from x-ray films. Hydrolisis of the protein bound of the chain in the textile and used many more indüstries field.In our study, the effect of various parameters on the protease from the Bacillus licheniformis with the SSF method was examined. SSF medium; rice husk, wheat bran, rice stalk, wheat stalk and barley husk were used as solid substrate. The best result was gained with rice stalk, therefore, rice stalk was used as solid substrate in the following studies. The suitable incubation time for the obtaining, for the protease has been determined as 72 th hour. The effect of substrat particle size on the protease production was tested. Maximum protease production was detected at 1500 µm substrat particle size.The effect of pH and temperature on the protease activity was tested. Maximum protease activity was detected at pH 9.5 and 50 oC respectivelly. To examine the effect of detergents on the protease activity, 1-5% SDS and Tween 40 were used. All results obtained were found to be lower than the control.İn studies done for the effect of planting amount and moisture level on the enzyme production, for protease has been determine suitable planting amount as %25 and suitable moiture level as %50.The effect of different nitrogen and carbon sources in %1 ratio on the production of enzyme was examined. While the best nitrogen sources for protease was detected as casaminoacid, urea and amonyum nitrate; the best carbon sources for protease was detected as sucrose, maltose and glucose.Key words: Bacillus licheniformis, protease, SSF
