Abstract (EN)

The usages of enzymes are used in biotechnology, industry, food, textil, paper, medicine and pharmaceutical. Enzymes obtained from microorganisms are produced by microorganisms have some advantages when compared with enzymes produced by plants or animals, they have considerably higher catalytic activity, they don?t from undesirable by-products, they are more stable and relatively cheap, and they can be obtained much quantity. Proteases that are a group of industrial enzymes have an important role. Proteases were used detergent, pharmaceutical, leather, meat, milk, beer indüstries, hydrolysis of proteins such as horn, feather and hair, recovery of silver from x-ray films and used many more industries field.In our study, the effect of various parameters on protease from the Bacillus licheniformis ATCC 14580 with the Solid state fermentation (SSF) method was evaluated. Rice husk, wheat husk, barley husk, cotton stalk, crushed maize and lentil husk were used as solid substrate for SSF medium. The best result was gained crushed maize, therefore, crushed maize was used as solid substrate in the following studies. The suitable incubation time for the obtaining, for the protease has been determined as 24th hour. The effect of substrat particle size on the protease production was tested. Maximum protease production was detected at 1500 ?m substrat particle size. Maximum protease pruductıon was detected at pH 8.0 and 37°C respectivelly. Inoculum volume was found to be 2 ml. The effect of carbon and nitrogen sources on protease pruduct were observed the highest with yeast extract and %1 galactose. The effect amount of solid substrate and mixed solid substrate on highest protease activity was investigated. The highest protease activity was observed at %30 mixed substrate (%25 Crushed maize+%5 Rice husk).
