?-amylase production from Bacillus subtilis ATCC 6051 wiith solid state fermentation (SSF)

Amylase are ona of the most important and great significance in the present industry of biotechonology. Amylase have several different industrial applications including foods, pharmacations, detergents, textile, and animal foods.The main goal of the present study is to realize the optimization parameters of the ?-amylase obtained from Bacillus subtilis ATCC 6051 by solid phase fermentation method.The best incubation temperature, inkübation time and pH were determined. Op timum enzyme activitiy. Enzyme showed optimum activity at 50 oC and ph 6.5. Amount of innoculum and initial moisture content suitable for enzyme production were found to be 30 % and 20 % respectively. The tap water was the best exraction medium. The best shaking speed for enzyme production was determined as 150 rpm. The effect of nitrogen one carbon sources on the enzyme production was examined. Ammonium sulfate, ammonium nitrate, ammonium chloride and malt extract were increased the production of enzyme, but cazein,pepton and tripton were reduced the enzyme production. Fruktose as a carbon sources was enhanced the enzyme inhibited the enzyme production by 55 %. The effect of metal ions on enzyme production was also examined. It was observed that MgSO4.7H2O, CaCl2 and FeSO4.7H2O were caused on increase in the enzyme production but ZnSO4.7H2O and CuSO4.5H2O were strongly inhibited the enzyme production.Keys words: Bacillus subtilis, ?-Amylase, Wheat Bran, Biotechonology, SSF
