Abstract (EN)
In this research, Cm-AAT antisense genes responsible for synthesis of aroma volatiles in melon (Cucumis melo L., var cantalupensis Naud) were transformed by Agrobacterium tumefaciens. Commercial recombinant RNAi - pFGC5941 plasmid transformed from Escherichia coli (DH5?) to Agrobacterium tumefaciens, was used.Based on the results, from the 1200 seeds used in the thesis 60 putative transgenic plants for AAT 1 gene, 39 putative transgenic plants for AAT3, 28 putative transgenic plants for AAT4 were obtained. These results were confirmed by PCR analysis. Transformation efficiency of antisens genes was found %2,5 for AAT1, %1,7 for AAT3 and %1,25 for AAT4 Cotyledone and proximal explants from the seeds were cultured on the selective medium. Proximal explants gave better results in view of regeneration and shoot rates for all genes. These results were confirmed by statistical analysis.
