Effect of rifampin in combination with allogeneic, alloplastic, and heterogenous bone grafts on bone regeneration in rat tibial bone defects

We aimed to evaluate different type of bone grafts (allogeneic, heterogenous, alloplastic) combination with rifamycin on healing of the bone defects and also compare the values of bone union, spongiosa, cortex and bone marrow in this study.Twenty-eight male Wistar albino rats weighting 250 to 400 gr were used in the treatment. The animals were divided 4 experimental groups. Thr animals were anesthetized with intraperitoneal injection of Ketamine (KetalarR, Eczacıbaşı, Turkey) and Xylazine HCL (RompunR, Bayer, Turkey). Prior to surgery, the skin was shaved and scrubbed with an antiseptic solution (1% iodine). Then bone cavities which were 10 mm long, 3 mm deep and 2 mm wide were formed by round burs with low rpm, under irrigation with sterile saline solution and standardised with the help of wooden models of the same dimensions on the tibias of the rats. In group 1, the right bone defect was irrigated with rifamycin (Rif, Kocak Farma, Turkey), the left bone defect was irrigated with sterile saline solution. In group 2, the right surgical defect was filled with rifamycin and allogenic bone graft (Raptos, Citagenix, Canada), the left defect was filled with allogenic bone graft alone. In group 3, the right experimental bone defect was treated with rifamycin and alloplastic bone graft (4Bone, MIS, Fransa), the left defect was filled with alloplastic bone graft alone. In group 4, the right bone defect was filled with rifamycin and heterogeneous bone graft (Bio-OssR, Geistlich Farma AG, Switzerland), the left bone defect was filled with heterogeneous bone graft alone. The rats were sacrified to assess of bone healing response at the postoperatively 21st day. The values of bone union, spongiosa, cortex and bone marrow were investigated in sections. The results were analyzed statiscally with Mann-Whitney-U test.In conclusion the histhopathological results revealed that the rifamycin only group have shown higher scores than control group and it was found statistically significant. There was no sign of bone healing in the rifamycin+ allogeneic bone graft group. There were similar clinical bone healing scores in the rifamycin+alloplastic group and rifamycin+heterogenous bone graft group when compared with control groups and no significant statistically differences were observed between these groups.Twenty-eight male Wistar albino rats weighting 250 to 400 gr were used in the treatment.
