Comparative diagnosis of caseous lymphadenitis in sheep by molecular and ELİSA method

Abstract (EN)

Aim: This work; Diagnosis of Corynebacterium pseudotuberculosis (C. pseudotuberculosis), which is the etiological agent of Caseous Lymphadenitis (CLA) in sheep in Diyarbakir region, using Polymerase Chain Reaction (PCR), Enzyme-Linked Immuno Sorbent Assay (ELISA) and bacteriological culture methods, isolation findings and tests. It was aimed to determine the specificities and sensitivities of the disease and thus to demonstrate their usability in the routine diagnosis of the disease. Material and Methods: The study material consisted of 2650 sheep in the central and surrounding districts of Diyarbakır. 90 sheep with clinically one or more abscessed superficial lymph nodes were selected. Swap samples were taken from blood samples and abscess contents. For the diagnosis of the agent, bacteriological culture and PCR analyzes were performed on swap samples and ELISA analyzes on blood samples. Results: According to PCR analysis of sheep in Diyarbakır region 26 (28,8%) positive, 64 ( 71.1% ) negative, ELISA analysis 17 (18,8% ) positive, 73 (81,1% ) negative and while 12 (13.3%) C. pseudotuberculosis was isolated in bacteriological cultures, staph of 16 were isolated from streptococ in 8 and e.coli in 6 of them. It was determined that 48 (53.3%) negativities were detected. Conclusion: It was concluded that PCR in clinical samples was more efficient, reproducible and faster than ELISA and microbiological culture in confirming C. pseudotuberculosis. It was revealed that caseous lymphadenitis infection was present in sheep in Diyarbakir region, and 28,8% positivity was detected in sheep with clinical symptoms according to PCR analysis.
