Propogation of sea daffodil (Pancratium maritimum) via organogenesis and microbulb methods

Abstract (EN)

In this study, it was carried out five treatments in order to determine best conditions for in vitro studies; 1) disenfenction method for seeds, which are starting material 2) In vitro seed germination medium 3) In vitro bulbous formation medium 4) Callus induction medium and explant type 5) Plant regeneration medium. The most appropriate method for seed disinfection was determined as %20 sodium hipoklorit hold for 15 min. The most suitable medium for seed germination and plant growth was found to be MS (Murashige-Skoog) without hormone. In the study, for the purpose of in vitro bulbous formation of plantlets, it was used medium which containing sucrose (20, 40 and 80 gl-1), BA (6-benzylaminopurine) (0, 1 and 2 mgl-1) and 2,4-D (2,4-Dichlorphenoxyacetic acid) (0, 0.1 and 0.2 mgl-1). In case of number of plants that formed bulbous and onion formation rate, all medium has been succesful. Diameter of bulb increased with high sucrose (80 gl-1) in growth medium. In terms of bulb weight, effect of plant growth regultors has been more ronounced in nutrient medium which containing low sucrose (20 gl-1). To callus induction, it was used mediums (including only 2,4-D and BAP (6-Benzylaminopurine) + 2,4-D) and explants (embryo, root, leaf). As a consequence of that, embryo explants and medium containing together 2,4-D (1, 2 ve 4 mgl-1) and BAP (1 mgl-1) has been successful. In order to shoot regeneration from callus, both mediums which containing BAP (2 mgl-1) and 2,4-D (0.5 and 0.2 mgl-1) has been succesful (% 90).
