Abstract (EN)
Wound dress which has largest share of medical textiles, is one of the significant applications. Studies carried out for wound healing purposes date back to ancient years. Firstly, plants were used and synthetic antioxidants replaced those. Although it has been used for many years, Pine bark has recently a wide area of application in nutrition health and medicine. The aim of this thesis is to design wound dressings using the extracts of Pinus brutia known as Turkish Pine, and to determine the wound healing performances of the obtained products in vivo experiments. In the study, Pinus brutia extracts were obtained using modern extraction techniques such as microwave and ultrasonic extraction apart from the traditional soxhlet extraction method. Optimum extraction conditions obtained using with Box-Bhenken design were elicited as 70ºC, 10 min., 900 W which yielded 987.32 mg gallic acid equivalent (GAE), 23.7 mg quercetin/g extract and 86.2% free radical scavenging capacity for microwave-assisted water/ethylacetate extraction; while 70 ºC, 20 min. and %50 power which yielded 811.84 mg gallic acid equivalent (GAE), 30.1 mg quercetin/g extract and 90.8% free radical scavenging capacity for ultrasonically-assisted water-ethylacetate extraction. Total phenol result of microwave ethanol extract (1083.5 mg/g extract), ultrasound ethanol extract (1014.8 mg/g extract) and soxhlet ethanol extract (813.9 mg/g extract) performed better than water/ethylacetate extract. Moreover, total flavonoid concentration showed that the highest value was attained with microwave (99.4 mg quercetin/g extract) while ethanol was used as solvent. After the extraction, the sodium alginate solution was prepared using Pinus Brutia extract and the gel was formed on the fabric by treatment CaCl2 solution to obtain wound dress. Wound healing performances of the wound dressings obtained using with extracts were tested on rats. At the end of the first week, 75.7 percent wound area of rats which treated with wound dress containing Pinus brutia gel, was healed, while in control group rats 48.6 percent wound area was healed.
