Microencapsulation of Lactobacillus acidophilus KPb4B strain by different methods and determination of their effects on bacterial viability by response surface methodology

In this study, in the light of information obtained from previous researches, Lactobacillus acidophilus KPb4b strains, the potantial probiotic properties of which were identified in the culture collection of Food Microbiology Research Laboratories Department of Faculty of Engineering and Architecture of Bolu Abant İzzet Baysal University and confirmed by molecular genetic methods, were used. In the scope of this study, microencapsulation was carried out by the emulsion technique of potential strain, electrostatic vibration / dripping method and double layer coating (second layer coating with chitosan on the first coating performed by the electrostatic vibration / dripping method). At the end of the study, it was tried to determine the appropriate parameters to be used to protect the viability of microorganism by using the response surface methodology. According to the results of the study, the difference in the number of viable microorganisms at the beginning and at the end of the analysis was low, indicating that the resistance of the bacteria to the analysis conditions was high. According to the results of the study, the effect of aljinat concentration was very important (p<0.01), and the interaction effect of aljinat concentration and chitosan concentration) was also very important (p<0.01). According to the results of the study, it was concluded that the alginate concentration and calcium chloride molarity in the coating and double layer (electrostatic vibration/dripping method + kitosan) coating methods significantly influenced the microencapsul size as a consequence of (p<0.01) and besides it was detected that aljinat concentration was significantly effective on the size (p<0.05) in the emulsion technique.
