Investigation of the effects of cell free extract from lactic acid bacteria on growth and biogenic amine production of food-borne pathogens

In this study, the functions of cell free (CF) extract from lactic acid bacteria (LAB) on the growth and biogenic amine (BA) production of food borne pathogens (FBP) in two amino acid (histidine and tyrosine) decarboxylase broth were investigated. The antimicrobial functions of CF extract on the growth of FBP were tested using agar diffusion method. Four LAB (Leuconostoc mesenterodies subsp. cremoris, Pediococcus acidophilus, Lc. lactis subsp. lactis, Streptococcus thermophilus and four pathogenic bacteria strains (Listeria monocytogenes, Staphylococcus aureus, Escherichia coli and Salmonella parathypi A were used. The result of the study showed that CF extract significantly inhibited growth of FBP (P<0.05). The highest inhibition effects were found by Strep. thermophilus for Staph. aureus, by Leu. mes. subsp. cremoris and Lc. lactis subsp. lactis for S. paratyphi A (12.33 mm inhibition zone diameter). There was significant differences in ammonia and BA production among the bacteria species (P<0.05). In histidine decarboxylase broth (HDB), histamine production by S. paratyphi A, L. monocytogenes, Staph. aureus and E. coli were 10387.54, 722.01, 7330.66 and 11163.53 mg/L, respectively. In tyrosine decarboxylase broth (TDB), tyramine production by these bacteria ranged from 367.54 to 5955.86 mg/L. CF extract from LAB generally inhibited AMN and BA production by FBP (P<0.05), although the functions of CF extract varied depending on not only decarboxylase broth but also bacteria strains. However, CF extract from LAB had stimulation effect on tyramine production in HDB and dopamine and serotonin production in TDB by FBP.
