Investigation of the effect of chalcon-imide derivative on surgical wound healing in rats undergoing laparotomy

Wound healing is the response of the organism to repair the wound caused by a traumatic injury. In this study, it was aimed to evaluate the effect of newly synthesized chalcone-imide derivative on wound healing in rats undergoing laparotomy. For this purpose, 48 healthy male wistar albino rats were used for the study. In the experiment, no experimental procedure was applied to the control group. The 1st and 2nd sham groups were given 1% DMSO by IP and the rats were sacrificed on the 7th and 14th days, respectively. In other groups, the abdominal wall was opened by laparotomy and 25-50 mg/kg-calcone was administered and sacrificed at the end of the 7th and 14th days. Total Antioxidant Level and Total Oxidant Level were measured in the serum of rats. MDA level, SOD and GSH-Px enzyme activity were studied from peritoneal tissue. Tissue and serum levels of IGF-I, EGF, FGF, TGF-β were determined by ELISA method. Tissue samples were evaluated histologically by immunohistochemical staining method. When we compare the serum protein MDA, SOD, GSH-PX levels of the groups; group-2 and group-3 GSH-PX serum levels were significantly higher than group-4 GSH-PX serum levels (p<0.001 and p<0.05) respectively. When the levels of tissue protein MDA, SOD, GSH-PX were compared; group 2 and group 3 tissue MDA values were found to be significantly lower than sham-2 group (P<0.05). Group-4 tissue GSH-PX values were found to be significantly lower than sham-2 and group-2 tissue GSH-PX values (p<0.001). A significant difference was found between the tissue TGF-β, IGF, FGF and EGF levels of the groups (p<0.05). As a result, chalcone application in wound healing increases oxidative stress and systemic inflammation and significant changes occur in antioxidant capacity in response to excessive formation of free oxygen radicals. It was found that chalcone derivative positively altered early wound healing in rats undergoing laparotomy by increasing growth factors and antioxidant capacity.
