Abstract (EN)

ABSTRACT In Leishmania Specimens, Total DNA Isolation, Purification and determination of Restriction Endonuclease Profiles In Çukurova and Şanlıurfa regions, in spite of the insecticides used against to Mosquito and Phlebotomus, cutaneous Leishmaniasis is spreading day after day. As a result of resistance in Leishmania sp. the struggle against these agents has been difficult and new drugs are required. The strugle has not only been difficult but also time consuming. It has always been possible that cutaneous leishmaniais can transform into viscerotropic. The goal of this study was the identification of genotypic differences between isolates and try to find out weather there are similarities genotypically with reference strains or not. Samples (25) were collected from patients who were effected by cutaneous Leishmaniasis in Çukurova and Şanlıurfa domains. Total DNAs were isolated from these samples and referans strains, the total DNAs of which were digested with EcoRI, Hindlll, Alul, Hinfl, Haelll and Rsal restriction endonucleases. The restriction fragment profiles of isolates were compared with each other and, also, with others of 14 referans strains, According to the electrophoretic results obtained in this study, genotypic heterogenities were observed between Çukurova and Şanlıurfa isolates and, also, in each region itself. In the comparision of DNA fragments of reference and isolates it was observed that there may be slight similarities between but never an exact one at all.
