The preventive role of lycopene and genistein in thioacetamide induced liver cirrhosis

ABSTRACTTHE PREVENTIVE ROLE OF LYCOPENE AND GENISTEIN IN THIOACETAMIDE INDUCED LIVER CIRRHOSISIn this study we investigated the protective effect of two antioxidant andanti-inflammatory nutrients, lcyopene and genistein, against TAA induced liver cirrhosis.Thirty five Wistar Albino rats were used and equally divided into 5 groups. The first group was control group. The other four groups were injected with intraperitoneal TAA 2 mg/kg for 8 weeks. The sekond was injected TAA alone. The third group received TAA+lycopene (6 mg/kg p.o.), the foruth grup received TAA+gensitein (1 mg/kg p.o.) and the fifth group received TAA+lycopene (6 mg/kg p.o.)+genistein (1 mg/kg p.o.) for 8 weeks. At the end of eight weeks animals were killed. Plasma ALT, AST, GGT, LDH, GSH-Px and tissue GSH-Px, TNF-?, TGF-ß, NFk-B, MDA and collagene type 1 were studied. Histological analysis for fibrosis and Immunohistochemical staining for ?-SMA were performed.In TAA group levels of serum ALT, AST, GGT, LDH and tissue TNF-?, TGF-ß, MDA, NF-Kb, type 1 collagene were significantly elevated and levels of tissue GSH-Px decreased compared with the control group. In group 3, 4, 5 levels of serum ALT, AST, GGT, LDHand tissue TNF-?, TGF-ß, MDA, NF-kB, type 1 collagene were significantly decreased and levels of tissue GSH-Px were elevated.In group 2 tissue fibrosis and ?-SMA expression significantly increased compared with control group (p?0.001). In group 3, 4, 5 tissue fibrosis and ?-SMA expression significantly decrased compared with group 2 (p?0.001). Tissue fibrosis and ?-SMA experssion were not significantly different in group 4 and 5 but, in both groups these parameters were significantly lower than group 3.In conclusion, lycopene and genistein are protective against liver cirrhosis via diminishing oxidative stress and inflammation. Genistein and genistein+lycopene combination are more potant than lycopene against liver cirrhosis.Key words: Liver cirrhosis, thioacetamide, lycopene, gensitein.
