EFFECTS OF DEXAMETHASONE ON THE KIDNEY IN THE LİPOPOLYSACCHARİDE-INDUCED MOUSE SEPTIC SHOCK MODEL Sepsis, which causes multiple organ failure, is the most common cause of mortality and morbidity in intensive care units. Excessive immune response caused by lipopolysaccharide in sepsis causes hypovolemia, shock and multi-organ failure. Our aim in this study is that we think it may be effective in acute kidney injury, which is a part of multi-organ failure that occurs in sepsis and causes high mortality; To see the effect of dexamethasone, which has anti-inflammatory properties. Our study was randomly divided into 3 groups of 21 male BALB-c mice (age: 8 weeks) after the approval of Fırat University ''Animal TestEthics Committee''. Control, sepsis and sepsis + dexamethasone (DEX) groups were formed. The control group was fed a basal diet, LPS + D-galactosamine + dimethylsulfoxide (10% DMSO: 0.5 ml / mouse) was administered to the sepsis group, LPS + D-galactosamine + dexamethasone (DEX) (6 mg / kg) was administered to the sepsis + DEX group. Renal function tests (BUN, creatinine) were studied in serum. Proinflammatory cytokines (TNFα, IL -6), oxidant parameter (MDA) and Sirtuin1 were studied from the kidney tissue. When the data of the control and sepsis + DEX groups were compared with the sepsis group data in terms of proinflammatory cytokines, MDA, Sirtuin1, BUN and creatinine levels, a statistically significant difference was found between all groups (p<0.001). A significant increase was observed in the levels of proinflammatory cytokines, MDA, BUN, and creatinine in the sepsis group compared to the healthy control group (p<0.001). A significant decrease was found in the levels of proinflammatory cytokines, MDA, BUN, and creatinine in the sepsis + DEX group compared with the sepsis group (p<0.001). While a significant decrease was observed in Sirtuin 1 levels in the sepsis group compared to the healthy control group, a significant increase was observed in the Sirtuin1 levels in the Sepsis + DEX group compared to the sepsis group (p<0.001). The results of this study show that dexamethasone has anti-inflammatory properties and a protective effect on sepsis-related kidney damage.
