Using of Leukocyte and Platelet Rich Fibrin as a Barrier Membrane and Evaluation Its Effect on Bone Regeneration The aim of this study was to compare the effects of Leukocyte and Platelet Rich Fibrin (PRF) and low-level-laser applied PRF (LLL-PRF) as barrier membranes with collagen membranes on new bone formation for the treatment of critical size bone defects. In this study, 5 male sheep with average weight of 40 kg and 18-24 months old were used. 4 tubes of 10 cc blood were drawn from the sheep and centrifuged at 2700 rpm for 12 minutes to obtain PRF. The right iliac crest of sheep was exposed by the mucoperiosteal elevation under general anesthesia. A total of 8 critical defects were created in each sheep. A deproteinized bovine bone graft was placed in all defects. The defects were divided into four groups with two defects from each group. The defects were covered respectively with collagen membrane or PRF in membrane form or LLL-PRF in membrane form. Membrane was not placed in one group which served as the control group. A total of 40 defects were obtained. Animals were sacrificed at the end of 12th week. The sections obtained were analyzed by histomorphometric method. According to the results of study, our main parametres which are the new bone formation (BV / TV), bone volume (BV) and bone surface (BS) rates were increased in all groups. In the comparison between the groups, the highest values in all main parameters were obtained in the collagen group and this difference was statistically significant (p<0,05). Higher values were observed in the LLL-PRF group compared to the PRF and control groups, but there was no statistically significant difference between them (p>0,05). Although the PRF group was more successful than the control group, no statistically significant difference was found (p>0,05). The findings of the current study suggest that PRF and LLL-PRF may act as barrier membranes. However, further studies are needed to increase the efficiency of the PRF membrane.
