Aim: To determine the frequency of Kleinefelter Syndrome with buccal smear method at 8-15 years old elementary school boys in Malatya and help their family about the treatment. Early treatment of those patients whose major part can not be diagnosed can prevent serious problems or make the interference possible. Therefore, we aimed to remind the forgotten and unused Barr body analysis method which is rapid, cheap and effective and to discuss to usability of Barr body analysis as a screening procedur.Materials and methods: Study was performed in April 2008 at Malatya centrum.4000 elementary school student whose schools were in different parts of the city was obtained. Buccal smear samples were taken from 400 students per school and no physical features were considered. Buccal smear samples were checked with microscope after appropriate painting method. With family approvals, the venous blood samples of the patients, whose smear was positive for Barr body, collected and chromosome analysis was done with GTG banding techniqueResults: Eight of 4000 school boys? smear sample were positive for Barr body. The chromosome analysis of these eight patients was reported as 47 XXY.The frequency of KS was 1/500 between 1-15 years school boys in Malatya.Conclusion: Although KS frequency that was established by buccal smear method was 1/500 in Malatya, when we think non detectable Kleinefelter mosaics and the children who can not be able to go school because of mental retardation, we can say that Kleinefelter Syndrome frequency is higher then our determination. Because of the low diagnosis rates, we consider screening all the male population before 11-12 ages with Barr body analysis.Key words: Klinefelter syndrome, 47, XXY, Barr body, infertility.
