Micropropagation of Maxma-14 sweet cherry rootstock on different medium and light intensities

Abstract (EN)

In this study, it was aimed to investigate and develop in vitro propagation methods using Maxma-14 cherry rootstock shoot tips of Maxma clones with Prunus avium L. x Prunus mahaleb L. hybrid. Effects of different concentrations of BAP (0.5, 1.0, 2.0 and 4.0 mgl-1), different BAP + GA3 combinations, light intensity at 4500, 6000 and 7500 lux during culture and shoot growth of WPM, MS, QL, NRM media it was investigated. The highest number of shoots in shoot growth was observed at a concentration of 1.0 mgl-1 BAP and three applications stand out in the combination of BAP and GA3. It was obtained with 6000 lux the highest number of shoots and shoot length beetwen the light intensities at different concentrations. In the study carried out with different media, it was found that shoots developed in MS and WPM media were more vivid and developed better. At the rooting stage of the study, it examines the shoot rooting of auxin types (NAA, IBA) added to MS and WPM media. The highest rooting rate (81 ± 10.1%) and number of roots (9.31 ± 1.44) were obtained in 1.0 mgl-1 NAA + WPM medium. it was observed that the roots growing in MS media were longer and thicker.
