A research of immunomodulatory effect on mesenchymal stem cells by flow cytometry and serology

Abstract (EN)

Aim: Fibroblast-like, adherent mesenchymal stem cells can differentiate to bone, cartilage and adipose tissues. They can be used for research or treatment purposes at the in vitro proliferations. Their properties of migration and establishment to several organs were shown in numerous animal and human organ transports. In addition to those, they have immunomodulatory properties and hematopoetic stem cell supportive activities. They were acquired currency in the treatments of tissue reject after autoimmune diseases and organ transports. The most successful results with mesenchymal stem cells were obtained at GVHD. At this study, the effects of lymphocyte subgroups and some cytokines on healthy mice which are mesenchymal stem cell transplanted were aimed to study.Patients and Methods: Mouse bone marrow stem cells were converted to mesenchymal stem cells by cell culturing method at the laboratory. CD3, CD4, CD19, CD34 and CD45 lymphocyte subgroups were studied with flow cytometry and IL-2, IL-3, IL10 and IL-15 levels were operated by ELISA method before and after the transplantation. 20 young and healthy mice were used at the study. Peripheric blood samples were obtained and analyzed before and after the transplantation.Results: At the comparison between before and after MSC transplantation, there was a significant difference at CD3 (p?0,0001) and CD45 (p?0,001), CD19 (p?0,05) levels, but there was no significant difference at CD4 (p?0,05) and CD34 (p?0,05). At the comparison between before MSC transplantation and normal saline infusion control group, there was a significant difference at CD45 (p?0,0001) and CD19 (p?0,05) levels, but there was no significant difference at CD3 (p?0,05), CD4 (p?0,05) and CD34 (p?0,05) levels. After MSC transplantation, there was a significant relation between IL-3 and IL-15 levels (p?0,0001) and there was a weak relation between IL-10 and IL-15 levels (p?0,05). At the comparision between normal saline infusion control group and after MSC transplantation, there was no significant difference at IL-10 (p?0,05) and IL-2 (p?0,05) levels, a statistically significant difference were found at IL-15 (p?0,001) and IL-3 (p?0,05) levels. By the help of the result obtained in this study, after the MSC transplantation on healthy mice, it has been shown that there might be some immune differences resulted in the immunomodulatory effects of MSCs.
