MiRNA – 370, 125B ve 23 a nin yüksek dereceli servikal i̇ntraepitelyal neoplazi ve skuamöz hücreli kanser olgularinda i̇fade düzeylerinin belirlenmesi

In this study, in order to determine the role of microRNAs in high-grade cervical intraepithelial neoplasia (HSIL) and cervical squamous cell cancer (SCC), to reveal a prognostic marker that allows individualization of diagnosis and treatment, it was used in tissue samples obtained from formalin-fixed paraffin-embedded blocks (FFPE). We aimed to determine the expression levels of miRNA 370, miRNA 125b and miRNA 23a and compare them with prognostic variables. In our study, patients who were operated due to cervical pathology in our clinic between 01.01.2006 and 31.12.2015 and whose pathology reports were SCC, HSIL and normal cervical tissue in microscopic examination performed by at least two pathologists were included in the study. miRNA was isolated from the sections obtained from the paraffin blocks and the expression levels were measured and compared using Real Time Qrt-PCR method. Then, these expression levels were compared with the prognostic variables of the disease. Shaphiro Wilk test and Skewness values were used for the control of the normal distribution of numerical variables in statistical analysis. ANOVA and Tukey tests were used in comparisons of 3 independent groups for variables with normal distribution, and Kruskal Wallis H test for non-normally distributed variables. Log Rank, Breslow, and Tarone-Ware tests were used to analyze the survival rates of the patients. Cox regression model was created with the Enter method to determine the independent variables affecting death. SPSS 25.0 package program was used for statistical analysis, P <0.05 was considered statistically significant. We found that miRNA 370, miRNA 125B and miRNA 23A were in statistically different distribution range in all three (3) groups, namely HSIL, SCC and Control group. It was found that miRNA 23-A increased in HSIL and SCC groups statistically. However, when expression levels of miRNA 370, miRNA 125B and miRNA 23-A were compared, no significant correlation was found between prognostic variables in patient groups.
