Abstract (EN)

Purpose: In this study, it was aimed to determine the in vitro regeneration abilities of Tulip by leaf explants culture Material and Method: In the study, in vitro regeneration created was tried as a result of in vitro leaves culture obtained from sterilized tulip bulbs and leaves obtained from tulip bulbs germinated in pots by sterilizing them in in vitro culture. For this aim the MS medium containing the hormones of 2,4-D (2 mg/lt), TDZ (2 mg/lt), Kinetin (2 mg/lt), NAA (2 mg/lt), 2,4-D+Kinetin (1 mg/lt), 2,4 for callus formation 2,4-D+Kinetin (2 mg/lt), IAA+TDZ (1 mg/lt), IAA+TDZ (2 mg/lt), IAA+BAP (1 mg/lt), IAA+BAP (2 mg/lt), NAA+Kinetin (1 mg/lt), NAA+ Kinetin (2 mg/lt), NAA+TDZ (1 mg/lt), NAA+TDZ (2 mg/lt), 2,4-D+TDZ (1 mg/lt) and 2,4-D+TDZ (2 mg/lt) were used. Results: While 6% callus was formed in MS medium containing 2,4-D (2 mg/lt) and NAA (2 mg/lt) hormone, 10% callus developed in media containing Kinetin (2mg/lt) hormone from the culture of leaves obtained by in vitro germination of Purple Prince Tulip bulbs. Conclusions: Callus regeneration was observed in media containing only 2,4-D (2 mg/lt), NAA (2 mg/lt) and Kinetin (2 mg/lt) hormones among the nutrient media subject to the study.
