Investigation of vasculogenesis and angiogenesis mechanisms in placentas of pregnancy resulting from normal and in vitro fertilization

Abstract (EN)

Objective: In this project, it was aimed to investigate the angiogenesis factors VEGF, VEGFR-2, CD34 proteins in term placentas obtained from pregnancies resulting from fresh embryo transfer and frozen embryo transfer. Method: The placentas obtained after delivery in pregnancies resulting from normal pregnancy and in vitro fertilization were collected and grouped. Some of the placentas were followed by routine light microscopy and histological sections were taken. Immunohistochemical staining was applied to these sections and the localization of the investigated proteins was determined. Necessary protocols for Western Blot experiments were performed with the other part of the tissues. Statistical analyzes were performed. Results: There was no statistical difference between the control group, DETP group and TETP group for VEGF protein amount. It was determined that the amount of VEGFR2 protein was statistically higher in the DETP group than the other groups. The amount of CD34 protein was found to be statistically higher in the control group than the other groups. As a result of immunohistochemical experiments, expression for VEGF protein was observed in syncytiotrophoblast, cytotrophoblast, villus vascular endothelial cells and stromal cells in all groups except villus stromal cells of TETP group. Expression of VEGFR2 protein was observed in syncytiotrophoblast, cytotrophoblast, villus vascular endothelial cells and stromal cells in all groups except cytotrophoblast cells of control group and syncytiotrophoblast of DETP group. For CD34 protein, expression was observed in syncytiotrophoblast, cytotrophoblast, villus vascular endothelial cells and stromal cells in all groups except the villus stromal cells and syncytiotrophoblast of control group. Conclusion: As a result of our findings, the expression amounts of the DETP group are higher in all of the VEGF, VEGFR2, CD34 proteins that we have investigated compared to the TETP group.
