Özet (EN)
Due to the heterogeneity of OCD and limited pharmacotherapy approaches designed for specific neurotransmitters, it cannot be promising for all patients. Therefore, in order to better understand the underlying mechanisms of this disease, it was aimed to investigate the possible relationship between OCD and proteins that differ significantly with the Liquid chromatography-mass spectrometry (LC-MS/MS) method. 14 patients diagnosed with Obsessive Compulsive Disorder according to DSM-5 criteria and 10 healthy controls were included in the study. Yale-Brown Obsession Compulsion Scale, Obsessive Beliefs Scale (SBS-44), and Metacognition Scale-30 were administered to the participants. For once, 10 cc peripheral blood was drawn from the participants into EDTA coated purple capped blood tubes. These blood samples were sent to the molecular biology laboratory in the Medical Biology Department for plasma isolation. The blood was centrifuged at 3500 rpm for 10 minutes at +40 °C and transferred to clean microeppendorf tubes with the supernatant containing the upper layer plasma. All isolated plasma samples were stored at -80° until the large-scale protein analysis LC-MS/MS based proteomics study. After the collection of all plasma samples, they were transferred to Istanbul Medipol University REMER Proteomics Unit in cold chain for proteomics study. In this study, the support of Bezmialem Vakif University Scientific Research and Projects (BAP) unit was used. When the differences between the results were evaluated in terms of both statistical significance and fold change, Plastin-2 (LCP1), Hook protein 1 (HOOK1), Apolipoprotein L1 (APOL1), Ig gamma-2 chain C region (IGHG2), Apolipoprotein D (APOD), Ceruloplasmin (CP) proteins showed a significant fold change (p ≤0.05, ≥1.4-fold change).
