Abstract (EN)

Abstract: Folliculogenesis begins in primordial germ cells and continues until mature follicle development. The result is a mature oocyte. The number of primordial follicles is high in the early embryonic period, but decreases until puberty, and the loss of approximately 1000 follicles per cycle ends with menopause. It is known that 99% of the follicles in the ovary undergo atresia and granulosa cells play a role in follicular atresia. It has been suggested that oxidative stress in granulosa cells has an initiating effect on atresia. Follicle development takes place under the control of hormonal and environmental stimuli. It is suggested that excessive use of Follicle Stimulating Hormone (FSH) impairs follicular development. FSH activates the expression of the phospho-mTOR (p-mTOR) pathway, inhibits the accumulation of microtubule-associated protein 1A/1B light chain 3 (LC3-I) 1.5 hours after administration and increases autophagy by inhibiting the p-mTOR pathway 3 hours later. is also known. voltage dependent anion selective channel protein 2 (VDAC2); associated with apoptosis. The presence of VDAC2 in dominant follicles has been shown, and it ensures the continuity of the follicle by protecting the follicles from apoptosis. Sirtuin-6 (SIRT6); It is known that inhibiting preantal follicle development prolongs the development of follicles and reduces ovarian aging. Materials and Methods: Mice were randomly divided into 4 groups with 6 mice in each group. After the mice were euthanized, their ovaries were collected. The ovaries were used to examine by immunohistochemistry and qPCR and were evaluated statistically. Results: Ovarian tissue and follicles were detected as a result of hematoxylin-eosin staining. With the follicle count, the number of healthy and atretic follicles was determined due to dose increase. Autophagic and mitochondral related markers and atretic follicles were detected by PCR support. Conclusion: In the study, whether high-dose gonadotropins commonly used in In Vitro Fertilization (IVF) treatments cause ovarian atresia; autophagic markers LC3B and SIRT6 and their mitochondrial relationship with VDAC2 were examined immunohistochemically and molecularly.
