Investigation of expressions of circadian rhythm genes in parafined embedded pancreas cancer tissue samples

Abstract (EN)
Objective: Circadian rhythm genes control important pathways in cells. Pancreatic cancer is one of the most difficult cancers to treat. The aim of this study was to investigate the differences in expression levels of circadian rhythm genes in pancreatic cancer tissues. Method: Total RNA was isolated from paraffin-embedded pancreatic tumor and normal tissues adjacent to tumor tissue and transformed into cDNA. BMAL1, CLOCK, PER1, PER2, CRY1, CRY2 expression levels of selected circadian rhythm genes were evaluated by performing reverse transcriptase chain reaction (RT-PCR) and quantitative real-time PCR (qRT-PCR). Expression levels were analyzed statistically and clinically. Results: Of the 22 patients with pancreatic cancer included in the study, 11 were female and 11 were male and their ages ranged from 43 to 85 years. The Wilcoxon test was applied in line with the 2-ΔΔCT values obtained as a result of the qPCR study performed in triplicate. Expression levels of selected genes in normal tissues adjacent to tumor and tumor tissue were compared. Accordingly, p = 0.529 (p> 0.05) for BMAL1, p = 0.538 (p> 0.05) for CLOCK, p = 0.947 (p> 0.05) for PER1, p = 0.827 (p> 0 for PER2), 05), p = 0.886 (p> 0.05) for CRY1, p = 0.938 (p> 0.05) for CRY2 values were reached. Conclusion: Tumor tissue of 22 patients with pancreatic ductal adenocarcinoma and normal tissue adjacent to tumor tissue were examined for differences in expression of circadian rhythm genes. When the average expression levels were compared in 22 patients, no statistically significant change was found. However, when each patient was examined individually, some changes in their expression were detected.
