Effect on pain of sodium and calcium channel inhibitors in parkinson rat model

Parkinson?s disease (PH), described as ?shaking palsy? by James Parkinson in 1817, is a neurodegenerative disease. Pain due to non-motor features is reported to affect 40-75% of the PD patient.Studies, especially in recent years, have revealed functional properties of ion channelsand their effects on homeostasis. Studies on calcium and sodium channels indicate that these channels are related with pain pathways. The aim of this thesis is to investigate effects on the formation of the pain and analgesia of impaired channel homeostasis in patients with Parkinson's disease.In this study, 50 rats were divided into 5 groups: control group, Parkinson's disease group (6-OHDA group), the PH group that was given jetokain, the PH group that was given verapamil and the PH group that was given jetokain+verapamil. An experimental model of Parkinson?s disease were induced via injection of 6-hydroxydopamine in all rats except control group. Effect drugs that were administred on pain sensation were evaluated by using tail-flick device in 3 weeks after surgical procedure. The values were statistically analyzed.When pain status before and after the surgical procedure in groups were compared with each other, the difference was statistically significant in all groups except control group. When measurements of pain, measured after treatment, were compared with each other in Parkinson's disease+jetokain, Parkinson's disease+verapamil and Parkinson's disease+jetokain+verapamil groups difference was statistically found significant in all these groups. However, when Parkinson's disease+jetokain+verapamil groups compared with Parkinson's disease+jetokain and Parkinson's disease+verapamil group, value of p for Parkinson's disease+jetokain+verapamil group had significantly higher (p=0.003) and the combination of these two channel blockers is performed more effective analgesia.According to these findings, analgesia can be achieved by using sodium and calcium channel blockers in PD. This analgesic effect of calcium channel blocker, verapamil, appeared to have more powerful than the sodium channel blocker, jetocaine.Key words: Pain, Jetokain, Parkinson's Disease, Verapamil
